摘要
目的初步探讨四君子汤总多糖对小鼠肠道黏膜体液免疫功能的影响。方法取NIH小鼠随机分为正常对照组、模型组、四君子汤总多糖(SJZPS)组和四君子汤去蛋白多糖(SJZFP)组,采用ELISA法检测小鼠小肠sIgA浓度,流式细胞(FCM)技术检测Peyer's结细胞表型,免疫组化法检测小鼠肠道TLR4的表达。结果模型组sIgA浓度小于正常对照组、SJZPS和SJZFP组,差异具有显著性(P<0.01);模型组CD19+细胞比例下降,与正常对照组比较,具有非常显著性差异(P<0.01),SJZPS和SJZFP组CD19+细胞比例上升,与模型组比较,具有非常显著性差异(P<0.01);模型组肠黏膜TLR4蛋白表达明显低于正常对照组,SJZPS和SJZFP组,具有非常显著性差异(P<0.01)。结论SJZPS和SJZFP可提高环磷酰胺抑制的体液免疫功能,具有肠道黏膜免疫调节作用。
Objective To preliminarily study effects of polysaccharide of Sijunzi Decoction (SD) on the function of intestinal mucosat humoral immune. Methods NIH mice were equally randomized into 4 groups: normal control group, model group, SD polysaccharide (SDPS) group, and SD free proteoglyean (SDFP) group. The intestinal sIgA content was detected by ELISA. The phynotype of Peyer' s nodel cells was examined with flow eytometer. Immunohistochemistry was used to detect the expression of TLR4 receptor in the intestine. Results Compared to the normal control group, SDPS and SDFP group, the concentration of sIgA was significantly decreased in the model group ( P 〈 0.01). Compared to the normal control group, the ratio of CD 19 positive lymphoeytes was significantly decreased in the model group ( P 〈 0.01 ). The ratio of CD19 positive lymphoeytes was significantly increased in SDPS and SDFP groups than that in the model group ( P 〈 0.01 ). The expression quantity of TLR4 was significantly decreased in model group than that in the normal control group, SDPS group and SDFP group ( P 〈 0.01). Conclusion SDPS and SDFP exert immunoregulation on the intestinal mueosal immune system.
