p53、Bcl-2、Bax、Bcl-xL和Bcl-xS蛋白在胰腺癌中的表达 被引量:7

Correlation of expression of apoptosis-related bcl-2-family(Bcl-2,Bax,Bcl-xL,Bcl-xS) and p53 in pancreatic cancer

摘要 目的探讨胰腺癌组织中p53和Bcl-2家族(Bcl-2、Bax、Bcl-xL、Bcl-xS)蛋白的表达及其与细胞凋亡的关系。方法免疫组化法检测35例胰腺癌组织中p53蛋白表达,分为p53阴性表达组(18例)和p53阳性表达组(17例);Westernblot法检测胰腺癌组织中p53、Bcl-2、Bax、Bcl-xL和Bcl-xS蛋白表达;TUNEL法检测35例胰腺癌组织中凋亡指数(AI)。结果Bcl-2蛋白在p53阳性表达组上调,而在p53阴性表达组下调(P=0.047);Bax和Bcl-xL蛋白在p53不同表达两组中都明显上调(P=0.274,P=0.334);Bcl-xS在p53阳性表达组明显下调,在p53阴性表达组明显上调(P=0.01);在p53不同表达两组中,AI分别为12.1±2.47和9.1±1.48(P=0.023);上述Bcl-2家族各成员表达与AI无相关性(P>0.05),而p53不同表达Bcl-2/Bax比值与AI有明显的相关性(P<0.01)。结论Bcl-2是依赖p53调节的抗凋亡蛋白,Bcl-xS是依赖p53调节的促凋亡蛋白,而p53主要通过调节Bcl-2/Bax比值发挥凋亡调节作用。 Objective The biological behavior of ductal adenocarcinoma of the pancreas is, in part, relevant to apoptosis, which is closely related to p53 and bel-2 family protein. A systematic investigation of the apoptosis-related bcl-2 family modulated by p53 in pancreatic cancer is lacking. The purpose of this study was to clarify the association between p53 and bel-2 family( Bel-2, Bax, Bcl-xL, Bcl-xS) expression in pancreatic cancer. Methods A total of thirty-five pancreatic cancer patients were studied. The expression of p53 protein in pancreatic cancer was assessed using the immunohistochemieal method, which categorized the pancreatic cancer patients into two groups: group 1, with immunonegative p53 (18 cases); and group 2, with immunopositive p53 (17 cases). The expression of p53, Bcl-2, Bax, Bcl-xL, and Bel-xS in the 35 pancreatic cancer eases detected by Western blotting was quantified with a densitometer. The apoptosis of the 35 pancreatic cancer eases was determined by terminal deoxynucleotidyltransferase-mediated UTP end-labeling(TUNEL). Results Bcl-2 was remarkably up-regulated in group 2( 15 of 17 ), but was down-regulated in group 1 (5 of 18 ). Bax was up-regulated in both groupl ( 15 of 18 ) and group 2 ( 15 of 17 ). Bcl-xL was up-regulated in both group 1 ( 14 of 18 ) and group 2 ( 15 of 17 ). Bcl-xS was remarkably down-regulated in group 2 compared to group l (P = 0. 01 ). The apoptosis indexes of groups 1 and group 2 were 12. 1 ± 2. 47 and 9. 1 ± 1.48, respectively( P = 0. 023 ) ,there was no relationship between AI and expression of Bel- 2, Bax, Bcl-xL and Bcl-xS, respectively( P 〉0. 05 ). AI was remarkably increased in high Bcl-2/Bax ratio group, and low in Bcl-2/ Bax ratio group( P = 0. 012). Conclusion Bcl-2 and Bel-xS represented the significant anti- and proapoptotic proteins, respectively, modulated through a p53-dependent pathway in pancreatic eancer, and p53 modulated apoptosis mainly through bcl-2/bax ratio.
