利用siRNA抑制环氧合酶-2对人胃癌裸鼠移植瘤的影响 被引量:4

摘要 目的观察环氧合酶2(COX-2)对人胃癌裸鼠移植瘤的影响并探讨其机制。方法构建靶向COX-2表达质粒和在BALB/c裸鼠皮下建立SGC-7901人胃癌细胞动物模型,用COX-2表达质粒基因转染治疗。结果构建的COX-2表达质粒在体内、外均可稳定表达。COX-2基因治疗可抑制裸鼠皮下肿瘤的生长和淋巴管的生成。COX-2基因转染下调血管内皮生长因子-C(VEGF-C)的表达。结论通过构建靶向COX-2 siRNA真核表达载体可抑制人胃癌裸鼠移植瘤的生长和淋巴管生成,COX-2对抑制胃癌细胞(SGC-7901)治疗有确切效果。
