摘要
目的探讨尼古丁对体外培养的人成骨样细胞(MG63)生物学性能的影响。方法用含不同浓度尼古丁(1×10-4mol/L和1×10-3mol/L)的DMEM高糖培养基体外培养MG63细胞,分别于第3、5、7、10、14天采用MTT比色法检测MG63细胞的增殖力;用Trizol提取总RNA,Real-Time PCR法检测MG63细胞骨桥蛋白、骨唾液磷酸蛋白的基因表达情况。结果与对照组相比,体外尼古丁处理对MG63细胞的增殖起抑制作用;高浓度尼古丁在第5、7天对细胞骨桥蛋白的基因表达起促进作用;并且尼古丁促进了骨唾液磷酸蛋白的基因表达。结论尼古丁可抑制成骨细胞的增殖,但不是通过抑制骨桥蛋白与骨唾液磷酸蛋白的分泌起作用。
Objective To evaluate the effects of nicotine on cell proliferation and the expression of extracellular matrix proteins( OPN and BSP) in the human osteosarcoma cell line MG63. Methods The cells were cultured with DMEM medium containing 10% fetal bovine serum with 0, 1 × 10^-4mol/L,and 1 × 10^-3mol/L nicotine for up to 14 days. The cell proliferation was evaluated by MTT. The expressions of OPN and BSP were estimated by determining the levels of their mRNAs using the Real-Time PCR. Results Nicotine pressed the cell proliferation and increased OPN expression in the presence of 1 × 10^-3moL/L nicotine after 5 and 7 days of culture. The amount of BSP increased during 14 days of culture with nicotine. Conclusion Nicotine suppresses osteogenesis of osteoblasts,but improves the mRNA of OPN and BSP.
