摘要 目的:研究实验性脑出血后大鼠神经细胞凋亡与NF-κB表达的动态变化,并初步探讨两者的关系。方法:将大鼠随机分为脑出血组、假手术组和对照组,应用立体定向技术将自体不凝血注入大鼠尾状核制备脑出血模型,大鼠分别在不同时间点断头取脑,连续冠状切片做TUNEL染色和NF-κB免疫组化染色。结果:凋亡细胞在脑出血后6h出现,1d后开始增多,3d达到高峰,并持续到7d仍有表达,并且与NF-κB的表达变化一致,各组与对照组比较,差异有统计学意义(P<0.05)。脑出血后TUNEL阳性细胞与NF-κB的表达呈正相关(r=0.957,P<0.01)。结论:脑出血后存在细胞凋亡,且可能与NF-κB的激活有关。 Objective: To study the dynamic change of the apoptosis of neurocytes and the expression of NF-κB after experimental intracerebral hemorrhage (ICH) in rats and investigate the relationship between them. Methods: The rats were randomly divided into ICH group, sham group and control group. ICH rat models were prepared with stereotactic infusion of autologous blood into caudate nucleus. The brains of rats were gotten off at different times and successive coronal slice was made and then stained. The apoptosis of neurocytes and the expression of NF-κB were detected respectively by TUNEL and immunohistochemistry staining. Results: Apoptosis cells appeared at 6 h after ICH, significantly increased after 1 d, reached peak at 3 d and continued to 7 d, and showed significant difference compared with control group (P〈0.05). The expression of NF-κB changed as the apoptosis. The number of TUNEL positive cells was positively correlated with the expression of NF-κB (r=0.957,P〈0.01). Conclusion: Apoptosis exists in the brain tissue after ICH and is related to the activation of NF-κB possibly.
