摘要
以茎尖为外植体,对海滨锦葵(Kosteletzkya virginica)再生体系的优化进行了研究,结果表明:茎尖萌发的最适培养基为MS+IAA0.3 mg.L-1+ZT0.3 mg.L-1,萌发率为91.11%;继代增殖最适培养基为MS+IAA0.1 mg.L-1+KT0.5 mg.L-1,增殖系数为4.67;生根培养基为MS(蔗糖3%、琼脂0.6%、pH值5.8),生根率为90%。炼苗移栽后,成活率可达60%。
We studied optimization of the regeneration system of Kosteletzkya virginica using meristem as the explant. The results showed that MS + IAA0.3 mg·L^-1 + ZT0.3 mg·L^-1 was the best culture medium for germination with a germination rate of 91.11% ; the best subculture medium was MS + IAA0.1 mg·L^-1 + ZT0.5 mg·L^-1, with a multiplication factor of 4.67 ; the best medium for rooting was MS (sucrose at 3 %, agar at 0.6 % and pH at 5.8) with a 90 % rooting rate. The survival rate of hardened - off seedlings was up to 60% after being transplanted.
