摘要
目的研究合欢皮不同路线提取的皂苷对小鼠移植瘤的影响。方法实验分模型对照组、4条不同路线提取的皂苷组〔5mg/(kg.d)〕,连续10d灌胃给药,多普勒彩色超声检测肿瘤血管密度,取材称重计算抑瘤率及器官指数。HE染色观察肿瘤组织形态学改变,TUNEL法测定细胞凋亡,免疫组化法检测CD31、HIF-1及FLK-1蛋白表达。结果与模型对照组相比,给药组小鼠肿瘤均被不同程度的抑制,组织形态发生改变,新生血管密度下降。肿瘤细胞凋亡,血管内皮细胞CD31和FLK-1表达下降。仅两条路线提取皂苷组分降低HIF的表达。结论4条不同提取路线提取的皂苷对小鼠肿瘤不同程度抑制,但其机制不尽相同,可能与其促进肿瘤细胞凋亡和抑制新生血管CD31和FLK-1表达有关。
OBJECTIVE To investigate the influence of Julibroside on the transplantable tumor of mice which were extracted by different methods. METHODS After intragastric administration of Julibroside for ten days, the microvessel density of the tumor was detected by color Doppler sonography.The inhibited tumor rate and the index of organ were calculated on mice. The morphologic change of tumor tissue was observed by HE staining. The apoptosis of tumor cells was detected by TUNEL method, and the protein expression of CD31, HIF-1 and FLK- 1 were measured by immunoeytochemical techniques. RESULTS Compared with the model group, the tumor of mice which was intragastrie administration with Julibroside was inhibited in various degrees. The morphouses of tumor tissue was changed, and the revascularizations were diminished. The apoptosis number of tumor cells was increased, and the expression of CD31 and FLK-1 were decreased, but the expression of HIF-1 was decreased by only two methods. CONCLUSION It shows that the tumor of mice which was intragastric administration with Julibroside was inhibited, but the mechanism was different. It maybe have something to do with the effect of in- creased the apoptosis number of tumor cells and of inhibited the expression of CD31 and Flk-1 of tumor vessel endothelial cell.
