摘要
以生长旺盛的侧金盏花嫩茎腋芽为材料,进行了芽的生长、分化及试管苗的生根、移栽、扦插和移植研究,建立起侧金盏花的无性系。结果证明:1/2MS+IAA0.5mg/L+NAA0.1mg/L是芽生长培养的理想培养基;MS+BA0.8mg/L+NAA0.3mg/L是生长芽分化培养和不定芽继代培养的理想培养基;1/2MS+NAA0.1 mg/L+IAA0.4 mg/L是不定芽生根培养和生根继代培养的理想培养基;炉灰渣是试管苗移栽和扦插的理想基质。试管苗移植后长势整齐、旺盛,各种生物学性状均与野生植株一致。
A study on growth, differentiation, rooting and transplanting of axillary buds were studied and establishment of asexual system of Adonis amurensis was also conducted. The results indicated that the ideal media for development of growing point was 1/2MS+IAA0.5mg· L^-1+NAA 0.1mg·L^-1. The best medium for differentiation was MS+6-BA0.8mg·L^-1+NAA0.3mg·L^-1. The medium of 1/2MS+NAA0.1mg. L^-1+IAA0.4mg· L^-1 was suitable for rooting. The cinder was the ideal matrix for plantlet culture with vigorous and variant characters.
