通心络抗家兔主动脉粥样硬化的作用及其机制 被引量:12

摘要 目的:观察通心络对主动脉粥样硬化家兔血清和动脉粥样硬化(AS)局部纤溶酶原激活物抑制物-1(PAI-1)及血管细胞黏附分子-1(VCAM-1)表达的影响,阐明通心络抗AS的可能机制。方法:24只大耳白兔随机分为常规饲料组、胆固醇饲料组和通心络组,饲养14周,检测实验前后血清胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL)、PAI-1、VCAM-1水平的变化;处死动物后取主动脉进行病理学检查;采用免疫组织化学方法测定AS局部PAI-1和VCAM-1阳性表达百分数。结果:实验前三组动物血清TC、TG、LDL、PAI-1和VCAM-1水平比较差异无显著性(P>0.05),14周后胆固醇饲料组和通心络组TC、TG、LDL、PAI-1和VCAM-1水平明显高于对照组(P<0.01),通心络组血清TC、TG、LDL、PAI-1和VCAM-1水平明显低于胆固醇饲料组(P<0.01);免疫组织化学检测胆固醇饲料组和通心络组AS局部PAI-1和VCAM-1的阳性表达百分数明显高于对照组(P<0.01),通心络组PAI-1和VCAM-1的阳性表达较胆固醇饲料组的表达明显减少(P<0.01)。结论:AS的发生伴有PAI-1和VCAM-1的过度表达,抑制PAI-1和VCAM-1的表达可能是通心络抗AS机制之一。 Objective To observe the effect of Tongxinluo on the expreeions of plasminogen activator inhibitor type- 1 (PAI-1) and vascular cell adhension molecule-1 (VCAM-1) in rabbits with atheroselerosis (AS), and explore the possible mechanism. Methods Twenty four rabbits were randomly divided into control, cholesterol diet, and Tongxinluo groups. The levels of serum cholesterol (TC), triacylglycerol (TG), low density lipoprotein (LDL), PAI-1 and VCAM-1 were measured before and after treatment. The aortas were harvested for pathologic morphological observation. Immunohistochemistry analysis was used to evaluate the positive percentages of PAI-1 and VCAM-1. Results There were no differences of TC, TG, LDL, PAI-1 and VCAM-1 between three groups before treatment (P〉0.05) . 14 weeks after treatment, the TC, TG, LDL, PAI-1 and VCAM-1 levels in cholesterol diet group and Tongxinluo group were significantly higher than those in control group (P〈0.01), while the TC, TG, LDL, PAl-land VCAM-1 levels in Tongxinluo groups were signficantly lower than those in cholesterol diet group (P〈0. 01) . The expressions of PAI-1 and VCAM -1 in aorta in cholesterol diet and Tongxinluo group were higher than those in control group (P〈0.01), and the expressions of PAl-1 and VCAM-1 in Tongxinluo group were lower than those in cholesterol group (P〈0.01). Conclusion PAI-1 and VCAM-1 over express in AS rabbits, while inhibiting the expressions of PAI-1 and VCAM is likely to be one of anti-AS mechanism of Tongxinluo.
