摘要
以山薯无菌苗茎段为试验材料,研究BA、NAA、JA、蔗糖浓度、光周期以及不同培养方式对试管零余子诱导形成的影响。结果表明,MS+2.0mg/LBA+0.1mg/LNAA+80g/L蔗糖+5.0g/L琼脂+1.5g/L活性炭,12h/d的光照时间(光照强度为2000lx)是试管零余子诱导较优的培养条件。经过2个月的培养,试管零余子诱导率达90%以上,零余子发生个数为1.44个/株。蔗糖浓度是影响试管零余子发生的最重要因素,高蔗糖浓度(80g/L)能促进试管零余子的发生。
Clean stem segments of Dioscorea fordii Prain et Burk were used as in vitro microcutting materials to induce formation of bulbils under mediums in the presence of various levels of BA, NAA, JA, sucrose and photoperiod. The results showed that the preferred condition for induction of in vitro bulbils was MS basal medium supplemented with 2.0 mg/L BA, 0.1 mg/L NAA, 80 g/L sucrose, 1.5 g/L activated coal and 5.0 g/L agar under a 12/12 h/h(day/night) photoperiod. For 2 months of culture, the stem segments had an in vitro bulbils inducing frequency of above 90%, and produced 1.44 bulbils/plantlet. The concentration of sucrose was the most important factor which affected the induction of in vitro bulbils, and higher concentration of sucrose(80 g/L) promoted the formation of in vitro bulbils.
