摘要
目的探讨针刺对家兔实验性动脉血管平滑肌细胞c—myc及c—foc基因表达的影响。方法家兔随机分为正常对照组(8只),再狭窄组(24只)。再狭窄组应用PTCA球囊导管对实验兔行双侧髂动脉内膜剥脱以建立动脉粥样硬化模型,术后1month髂动脉造影示50%以上狭窄者再行PTCA球囊扩张术,以建立再狭窄模型成功后,将造模动物随机分成模型组、针刺组、药物组,每组8只。自术前2d至术后14d为l疗程,针刺内关(双)、厥阴俞(双)、心俞(双)、膻中、足三里(双)等穴。标本石蜡切片,免疫组化,DAB显色,苏木素复染。观察各组血管平滑肌细胞c—myc、c—foc基因表达。结果模型组平滑肌细胞c—myc及c—foc基因表达明显高于正常组(P〈0.01);针刺组c—myc及e—foc基因表达明显低于模型组(P〈0.01)。结论针刺能抑制兔再狭窄血管平滑肌细胞增殖,抑制c—myc及c—fnc基因表达,调节平滑肌凋亡,可能是针刺预防动脉再狭窄的作用机制之一。
objective To observe the effects of acupuncture on the treating vascular smooth muscle cell and gene expression of c - myc, p53 in rabbits. Methods These rabbits were randomly divided into normal control (8 rabbits) and restructure (24 rabbits) groups. To establish artherosclerosis model applying FFCA saceulus proprius to denudation arteria iliaca inner membrane two sides in restructure group. One month after operation, PTCA sacculus proprius dilatation was made if arteria iliaca opacification indicates that there were artery stricture above 50 percent p after succeeding in establishing the restricture model, then the model animals were divided into model, acupuncture and probucol groups, 8 rabbits in every group. It is a course treatment form the first 2 days before operation to the 14th day after operation, acupuncturing aoupoints of Neiguan (pair), Jueyinshu (pair), Xinshu (pair}, Danzhong, Zusanli (pair) and so on. Preparation of paraffin section, immunity histochemistry , the coloration of DAB, the afterstrain of hematoxylin. Then the c - myc, e - foe gene expression of smooth musle cell in every group were observed. Results The c - mye and c - foc gene expressions of smooth muscle cell in model group were prior to those in the control group obviously (P 〈 0.01 ) ; The gene expressions of c - myc and c - foc in acupuncture group were lower than those in the model group (p 〈 0.01 ) Conclusion Acupuncture can restrain the hyperplasy of restenosis of vascular smooth muscle in the rabbits, restrain the gene expression of c - mye and c - foc and adjust the apoptosis of smooth muscle. This may be one of the mechanisms of preventing arterial restenosis with acupuncture.
