摘要
目的探讨兔VX2肿瘤超声造影定量新生血管血流灌注参数与肿瘤组织周细胞变化的关系。方法选用VX2肿瘤肌肉模型兔10只,应用超声造影连续脉冲对比(comparative pulse serial,CPS)技术及ACQ定量分析软件测定肿瘤血管血流灌注各参数值和峰值强度比率(rate of peak index,RPI),并与肿瘤组织标记周细胞的免疫组化染色检测的微血管密度(micro vascular density,MVD)和周细胞数(pericytes number,PN)对照,进行相关性分析。结果肿瘤组织造影血流灌注参数峰值强度(peak intensity,PI)为(15.29±4.14)dB,RPI为(2.95±1.26)dB/s;肿瘤组织标记周细胞的MVD为(17.75±7.60),PN为(26.10±10.59);肿瘤组织造影血流灌注参数PI与MVD和PN显著正相关(r值分别为0.866、0.857,P<0.01),RPI与MVD和PN显著正相关(r值分别为0.647、0.648,P<0.01),而到达时间(arrivaltime,AT)、达峰时间(time-to-peak,TTP)与MVD、PN无相关性。结论超声造影血流灌注参数PI和RPI可以间接反映肿瘤血管生成中周细胞的数量、分布、密度以及微血管密度等变化。
Objective To VX2 tumor at ultrasonography and c study the correlation between perfusion parameters of new blood vessels in hange of pericytes in tumor tissue of rabbits. Methods Ten rabbits with implanted VX2 tumor cells were enrolled in this study. Comparative pulse serial (CPS) technique and ACQ software were used to detect the ratio between perfusion parameters of tumor vessels and rate of peak index (RPI). Micro vascular density (MVD) was detected in tumor tissues stained with immunohistochemistry and compared with that in pericytes, and their correlation was analyzed. Results The mean PI and RPI in perfused tumor tissue were 15.29 ±4.14 dB/s and 2.95 ± 1.26 dB/s, respectively. The mean MVD and the number of pericytes in tumor tissue were 17.75 ±7.60 and 26.10 ± 10.59, respectively. The PI and RPI were positively correlated with MVD and pericytes. Conclusion Ultrasonic PI and RPI can indirectly reflect the changes of pericytes during the angiogenesis in tumors.
