摘要
目的检测癌-睾丸抗原SSX1-5 mRNA在视网膜母细胞瘤(RB)组织中表达情况,以及用于RB免疫治疗的可能性。方法采用逆转录-聚合酶链反应(RT-PCR)技术对18例RB组织、18例非肿瘤病变视网膜组织及18例眼部正常组织中SSX1-5 mRNA的表达进行分析。结果18例RB组织中,SSX1、SSX2和SSX4 mRNA的表达率分别为33.3%(6/18)、5.6%(1/18)和22.2%(4/18);未检测到SSX3和SSX5的表达。RB组织中,至少表达1个SSX基因者为50.0%(9/18);表达2个或2个以上者为11.1%(2/18)。在非肿瘤病变视网膜组织以及眼部正常组织中无1例表达SSX1-5 mRNA。SSX1-5基因的表达与年龄、性别、肿瘤大小、分化程度和临床分期等均无显著相关性(P〉0.05)。结论SSX1-5在RB组织中表达频率不尽相同,但具有较强的特异性,提示该基因家族有可能成为RB免疫治疗的靶抗原。
Objective To investigate the expression of cancer-testis antigens(CTA) SSX1-5 mRNA in retinoblastoma(RB) and the possibility of applying these antigens as targets for specific immunotherapy for RB.Methods Expression levels of CTA SSX1-5 mRNA were detected with reverse transcription polymerase chain reaction(RT-PCR) in 18 RB tissues,18 non-tumor retinal tissues and 18 normal eye tissues.Correlation between the expression of CTA SSX1-5 and clinical data was evaluated.Results Of 18 RB tissues,SSX1,SSX2 and SSX4 mRNA were expressed in 33.3%(6/18),5.6%(1/18) and 22.2%(4/18),respectively.About 50.0%(9/18) and 11.1%(2/18) of RB tissues positively expressed at least one and two members of SSX1-5,respectively.In addition,no samples of non-tumor retinal tissues and normal eye tissues expressed SSX1-5.No relationship was found between the expression of CTA and clinical indicators such as age,gender,tumor size,differentiation and clinical stage(P〉0.05).Conclusion CTA SSX1-5 are expressed with different percentage in RB,but with high specificity,which are possibly promising targets for antigen-specific immunotherapy of RB.
