系统性红斑狼疮患者外周血IL-17表达及其与疾病活动性的关系 被引量:17

Expression of interleukin-17 in peripheral blood of patients with systemic lupus erythematosus and its correlation with disease activity

摘要 目的研究系统性红斑狼疮(systemic lupus erythematosus,SLE)患者外周血白细胞介素-17(interleukin-17,IL-17)蛋白及基因的表达水平及其与疾病活动性的关系。方法用酶联免疫吸附试验法(ELISA)检测26例SLE患者及21例健康对照者血浆中IL-17的蛋白水平;以β-actin为内参照,采用荧光实时定量反转录聚合酶链式反应(Real-ti me RT-PCR)检测两组外周血中IL-17 mRNA表达水平。结果SLE组患者血浆IL-17含量明显高于健康对照组[(18.04±6.04)pg/mLvs(12.73±6.81)pg/mL,P<0.01];SLE组患者外周血IL-17 mRNA表达水平高于对照组;IL-17蛋白含量与mRNA表达水平呈正相关(r=0.7,P=0.007 0)。SLE组血浆IL-17水平与SLE疾病活动性指数(SLEDAI)积分和抗dsDNA抗体水平呈正相关(r=0.459,P<0.05;r=0.487,P<0.05);与血清C3水平呈负相关(r=-0.373,P<0.01)。结论SLE患者体内IL-17蛋白分泌及基因表达水平存在明显异常,两者呈正相关,且与疾病活动性有明显相关性,提示IL-17可能在SLE发病中起重要作用。 Objective To determine the protein and mRNA levels of interleukin-17(IL-17) in the peripheral blood of patients with systemic lupus erythematosus(SLE),and to analyze the relationship between IL-17 and disease activity. Methods Twenty-six hospitalized SLE patients and twenty-one normal controls were studied.Plasma protein and mRNA of IL-17 in peripheral blood were measured by ELISA and Real-time RT-PCR respectively. Results IL-17 level and its mRNA level increased in SLE patients compared with that in normal controls.Plasma concentration of IL-17 showed a significant positive correlation with SLEDAI score and anti-dsDNA antibody level,and a significant negative correlation with serum C3 level. Conclusions Plasma IL-17 protein and mRNA expression level in SLE patients increased significantly and had close relationship with disease activity,which might suggest that IL-17 play an important role in the pathogenesis of SLE.
