摘要 目的:研究大鼠嗅鞘细胞(OECs)表面免疫抗原分子的表达。方法:培养OECs,流式细胞仪检测其表面主要组织相容性抗原I、II类(MHC-I、MHC-II)分子表达情况,免疫细胞化学法检测髓鞘碱性蛋白(MBP)、髓鞘含脂质蛋白(PLP)、髓鞘结合糖蛋白(MAG)的表达情况。结果:OECs高表达MHC-I分子(78.4±4.45)%,中等表达MHC-II分子(43.2±2.98)%,不表达MBP、PLP、MAG等髓鞘蛋白。结论:OECs高表达的MHC分子可能诱导异体OECs移植的免疫反应,但MBP、PLP、MAG等自身抗原的缺如使其在自体移植治疗自身免疫性脱髓鞘病中有一定应用前景。 Objective: To investigate the immunogenicity of olfactory ensheathing cells (OECs). Methods: OECs were cultured and purified by a method based on the different rate of attachment of diverse cultured cells. Flow cytometry (FCM) was used to measure the expressions of major histocompatibility complex class Ⅰ and Ⅱ (MHC Ⅰ and MHC Ⅱ) and the immunocytochemistry was used to detect the expressions of myelin basic protein (MBP), myelin associated glycoprotein (MAG) and myelin proteolipid protein (PLP) on OECs. Results: OECs expressed a high level of MHC-Ⅰ (78.44±4.45)% and a moderate level of MHC-Ⅱ (43.2±2.98)% molecules but not any autoantigen molecule such as MBP, PLP or MAG. Conclusion: The high expression of MHC molecules on OECs indicates a possible allograft immune-response in the OECs transplantation. However, the lack of autoantigen molecules such as MBP,MAG or PLP suggests that these cells be in an immature state, which means a promising per- spective in autologous OECs transplantation in treatment of autoimmune disorders such as multiple sclerosis (MS).
