摘要
采用蛋白质组学方法,检测了产前注射维生素AD3E(V-AD3E)对围产期奶牛乳蛋白表达的影响.二维凝胶电泳分离了产后第1天和第21天乳蛋白组分,凝胶经染色、酶切、图像采集和斑点比对,与对照(不注射V-AD3E)组第1天乳蛋白图谱相比,注射组第1天乳蛋白图谱上有5个区域的蛋白表达上调,经高效液相色谱串联离子阱质谱对差异表达蛋白进行鉴定,主要为免疫球蛋白G(IgG)和白蛋白.产前注射V-AD3E上调了初乳中的IgG和白蛋白,不仅为饲喂初乳的犊牛提供了更好的免疫保护,而且提高了处于免疫抑制阶段母牛的抗病力.
The effects of vitamin AD3E (V-AD3E) solution on the milk protein expression of periparturient dairy cows were determined using proteomic analysis. Milk Protein from d 1 and d 21 after calving were separated and identified by two-dimensional electrophoresis combined with high performance liquid chromatography tandem ion trap spectrometry (HPLC/MS/MS). IgG and albumin in colostrum from dairy cows injected with V-AD3E solution were up-regulated compared with not injected ones. No changes of milk protein composition were observed from d 21 postpartum between injection and non-injection. Injecting V-AD3E solution into the periparturient dairy cows prior to calving up-regulated IgG and albumin levels in colostrum,which, besides transferring higher levels immune constituents to the offspring, fed the colostrum, also promotes development and digestive tract protection in the neonate, and defends the cows who are in immune suppression period against the invasion of pathogenic bacteria,fungi and virus.
