乙酰半胱氨酸对急性酒精性肝损伤大鼠NF-κB和TNF-α的影响 被引量:5

摘要 目的探讨乙酰半胱氨酸对酒精所致大鼠急性肝损伤的保护作用及机制。方法采用酒精灌胃法制作急性酒精性肝损伤大鼠模型。50只雄性Wistar大鼠随机分为5组(n=10):正常对照组、急性酒精肝损伤模型组、乙酰半胱氨酸低、中、高剂量(150,300,600mg/kg)组。测定并比较各组大鼠肝脏中核转录因子-κB(NF-κB)的表达及血清中肿瘤坏死因子-α(TNF-α)的含量。结果急性酒精性肝损伤大鼠模型制作成功。与模型组比较,治疗结束后,乙酰半胱氨酸各剂量组急性酒精性肝损伤大鼠肝组织中NF-κB的表达均减少(P<0.01),血清TNF-α的含量降低(P<0.01),炎性介质的释放不同程度地减少。结论乙酰半胱氨酸可以减轻肝脏的炎性损伤,对大鼠急性酒精性肝损伤起到保护作用。 Objective To investigate the protective effect of N-αcetylcysteine (NAC)on acute alcoholic liver injury in rats and its possible mechanism. Methods The liver injury models were induced by infusing alcohol into stomach of rats. Fifty male Wistar rats were randomly divided into 5 groups:control group, acute liver injury model group, and low, middle, high doses of N-acetylcysteine( 150,300, 600 mg/kg)groups. The expression of nuclear factor-kappa B(NF-κB) protein and tumor necrosis factor-α(TNF-α) content were assayed and compared between groups. Results The acute alcoholic liver injury models were established successfully. Compared with model group, the expression of NF-κB and serum TNF-α content significantly decreased in NAC groups(P 〈 0.01 ), and the release of inflammatory factors decreased to different degrees. Conclusion NAC can protect from acute alcoholic liver injury by alleviating the inflammatory injury.
