Study on preparation of extract from Radix Angelicae and its quality control

摘要 目的采用HPLC法同时测定当归提取物中阿魏酸和藁本内酯的含量,优化当归提取物的制备工艺。方法以阿魏酸和藁本内酯为指标成分,比较水提法、碱水提法、乙醇浸渍法、乙醇热回流法、乙醇醇渗滤法的优劣,确定制备当归提取物的工艺路线;采用L9(34)正交试验设计优化其提取工艺。结果阿魏酸0.112~1.125μg与峰面积线性良好(r=0.9999),平均回收率为99%(RSD=2.91%),藁本内酯1.368~13.680μg与峰面积线性良好(r=0.9999),平均回收率为104%(RSD=1.46%)。当归提取物的制备方法为乙醇渗滤法,最佳工艺条件:用70%乙醇浸渍24h,渗滤10倍量。结论乙醇渗滤法提取效率高,重复性好,可为当归流浸膏采用乙醇渗滤法制备提供依据;HPLC法可用于当归提取物中阿魏酸和藁本内酯的定量分析。 OBJECTIVE To optimize preparation process of extract from Radix Angelicae and establish a quality control method for the extract. METHODS HPLC method was used for determination of the content of ferulic acid and ligustilide simultaneously in the extract. Comparison experiments were carried out to choose extract methods for extract from Radix Angelicae. L9 ( 3^4 ) orthogonal experiment was used to optimize the extraction process of Radix Angelicae. RESULTS The content of ferulic acid presented a good linearity with the area of peak in the range of 0.112 - 1. 125 μg ( r = 0. 9999 ) and its average recovery rate was 99% with RSD of 2.91% ; The content of ligustilide presented a good linearity with the area of peak in the range of 1. 368 - 13. 680 μg ( r = 0. 9999) and its average recovery rate was 104% with RSD of 1.46%. The optimal method and conditions for extraction: coarse powder of Radix Angelicae was steeped for 24 h and then percolated with 10 times of 70% ethanol. CONCLUSION It was indicated that percolation is a optimal method for preparation of standard extract from Radix Angelicae. HPLC method can be used for quality control of the standard extract from Radix Angelicae.
