摘要
近些年来,国内外对血浆蛋白粉的利用已经取得了较大的进展,大都采用现代酶解技术,但水解率较低,水解不彻底。本文拟采用正交试验法筛选出最佳的蛋白水解酶,并结合碱水解方法利用单因素和多因素试验设计方法来优化蛋白酶—碱水解猪血浆蛋白的条件。得到最佳水解条件为:加入8000u/g的胰酶(加酶量以蛋白质含量计),55℃水解5h;之后加入8%的碱,80℃水解3h,达最佳水解条件,水解率达到77.1%。
In recent years, the utilization of plasma protein powder has made a a great progress at home and abroad. Most of them use enzymolysis technology. But the hydrolysis rate is low and not complete. This paper used orthogonal design to find the optimal proteolytic enzymes and combined with alkaline hydrolysis method using single factor and multi-factor experimental design methods to optimize the protease-alkaline hydrolysis conditions of the porcine plasma protein. The best hydrolysis conditions are: 8000u/g of trypsin (plus the amount of enzyme to total protein content), 55℃ hydrolysis for 5h; then add 8% of the alkali, 80℃ hydrolysis for 3h, under the best hydrolysis conditions, the hydrolysis rate reached 77.1%.
