摘要
采用响应面法对能够高效提高黏红酵母色素产量的发酵培养基进行优化。在前期单因素比较试验已筛选出蔗糖、蛋白胨、酵母提取物、接种量对黏红酵母色素产量影响显著的基础上,进行最陡爬坡实验逼近最佳响应面区域,最后通过Box-Behnken实验设计,利用SAS软件进行回归以及岭嵴分析得到各因素的最佳浓度。最终使色素产量可达16.56μg/mL,比初始培养基提高了约77%。
The pigments production of fermentation medium of Rhdotorular Glutinis was optimized by using the response surface methodology. Firstly, the influences of different single factor experiment were studied, It shows that fours factors playing the important roles in the sucrose, peptone, yeast extract powder and inoculating amount. The path of steepest ascent was used to approach the optimal region of the fermentation conditions subsequently. In the third step, the concentrations of those four main factors were further optimized by using Box-Behnken and Response Surface Analysis. By solving the quadratic regression model equation and ridge analysis, the pigments production increased 77% than before. It showed that Response Surface Methodoldgy was proper and a good choice for optimization.
