摘要
随着数十种模式生物基因组全序列测定的完成,生命科学的研究已经进入后基因组时代.在这个时代,由于全基因组的序列信息并不足以解释及推测细胞的各种生命现象,蛋白质才是细胞活性及功能的最终执行者,所以对核酸、蛋白质等生物大分子功能和相互作用的研究将会成为学术界的主流.体外展示技术作为一种新兴的多肽和蛋白质配体发现的重要工具,在基因和蛋白质的结构和功能研究方面有着不可或缺的作用.本文在对目前常用的几种体外展示技术的基本原理和应用进行综述的基础上,对其技术特点进行了比较,以期望能够帮助研究者根据不同的要求和目的选择适宜的方法.
Abstract With the sequencing of tens of model life-forms,researches on llfe science will consequently step into the age of post-genomics. In this age, sequence information is not enough to explain or predect all the phenomena taking place in the cells. It is proteins that perform as a major executive in determining the function and maintain the life of cells. Thus ,it will become a main stream for researchers to cencentrate on the interaction between such bio-molecules as nucleic acids and proteins. Serving as a new tool in finding the ligands of peptides and proteins, the in vitro display technology will play an indispensable role in researching the structures and functions of gene and proteins. Based on summarizing the priciples and applications of several in vitro display technologies, this review compared their technical characteristics, hoping that it can help researchers to select a proper method according to their special requires and purposes.
