氨基酸对大肠杆菌BL21产嘌呤核苷磷酸化酶的影响 被引量:2

摘要 采用单因素方差分析,考察了合成培养基中各种氨基酸对大肠杆菌BL21产嘌呤核苷磷酸化酶总酶活的影响;同时采用聚类分析和粗糙集分析,考察了复合培养基中氨基酸对大肠杆菌BL21产嘌呤核苷磷酸化酶总酶活的影响。结果表明,在合成培养基和复合培养基中对总酶活影响显著的是不同的氨基酸。在复合培养基中,丙氨酸和甘氨酸对产酶有明显促进作用且存在交互作用,两者浓度分别控制在80-140mg·L^-1和140-200mg·L^-1之间时能够很好地保证发酵产量。 The influence of amino acids in synthetic medium on total enzyme activity of purine nucleosidephosphorylase(PNPase) produced by E. coli BL21 were investigated by one-way ANOVA. Furthurmore, the effects of the same amino acids in the complex medium were studied by cluster analysis and rough set analysis. The major amino acids which affecting the total enzyme activity remarkably in the synthetic medium and the complex medium were obtained respectively. In the complex medium,it was found that adding alanine and aminoacetic acid could obviously improve the production of PNPase and there was interaction between them. When the alanine concentration was in 80-140 mg ·L^-1 and aminoacetic acid concentration was in 140-200 mg · L^-1 , the high yield of fermentation could be ensured.
