对小白鼠经不同途径注射抗原后脾单个核细胞的细胞因子IL-1βIL-2和IFN-γ的mRNA表达 被引量:6

摘要 应用RTPCR结合激光密度扫描定量分析方法,观察了不同途径免疫小白鼠的脾单个核细胞的细胞因子IL1β,IL2和IFNγ的mRNA动态表达水平。结果显示,穴位免疫组细胞因子IL1β和IL2的mRNA表达峰值和持续时间均大于非穴位免疫对照组。穴位免疫组与对照组比较,IFNγ的mRNA表达水平无统计学差异。这一结果在mRNA水平上证明经后海穴途径免疫可增强机体的免疫功能。提示在穴位免疫激活机体免疫应答的过程中,除了免疫系统自身调节外,尚有其他因素参与免疫调节。 At the present test a reverse transcription polymerase chain reaction (RT PCR) and a laser density scanning quantitative analysis method were used for observing the dynamic expressions of the mRNAs of cytokcines-IL 1 β, IL 2 and IFN γ in mononuclear cells of spleens of mcie immunized by various routes. The test result showed that the expression peak values and the lasting intervals of mRNAs of IL 1 β and IL 2 at the group of acupoint immunisation were larger and longer than that of the control group of non acupoint immunization but in comparison of the acupoint group with the control group their expression levels of mRNAs of IFN γ were non statistics different, which indicated that the immunity function of the mice immunized with antigens by a route of Houhai acupoint was enhanced at the level of mRNA and which suggested that in the progress of the body's immune response activated by the acupoint immunization, other factors took part in the immunoregulation in addition to the self regulation of the immunity system of the immunized mice.
