摘要
为探讨山东宁阳大枣组培快繁技术,以宁阳大枣新生枝条为外植体,通过诱导愈伤组织形成及不定芽分化,对建立高效的快繁体系进行研究。试验结果表明:同一基因型的不同取材部位其培养效果间差异不大;枣树新生枝条是建立无菌外植体的合适的外植体材料,适宜枣树分化愈伤组织的培养基是MS+BA1.0mg/L+KT1.0mg/L+NAA0.01mg/L+琼脂0.75%+蔗糖3%。适宜不定芽诱导的适宜培养基是MS+BA1.0mg/L+KT1.0mg/L+NAA0.05mg/L+琼脂0.75%+蔗糖3%。
To study the establishment of high efficient regeneration system of Ningyang Chinese date, its new shoots were used as explants to induce callus and differentiate adventitious buds. The results showed that new shoots of jujube were suitable for explants.MS＋BA1.0mg/L＋KT1.0 mg/L＋NAA0.01 mg/L was suitable for inducing callus; MS＋BA1.0mg/ L＋KT1.0 mg/L＋NAA0.05 mg/L was suitable for cluster buds.
