酶催化动力学光度法测定盐酸多巴胺

Determination of dopamine hydrochloride by enzymatic catalytic spectrophotometry

目的:建立了一种简单、灵敏的酶催化动力学光度法测定盐酸多巴胺的新方法。方法:在pH9.8的NH3-NH4Cl的缓冲溶液中,利用盐酸多巴胺对牛血红蛋白(Hemoglobin,Hb)模拟酶催化体系的抑制作用,研究了该抑制反应的最佳实验条件及动力学行为。结果:测定的线性范围为1.1×10-7～1.1×10-5mol.L-1,检出限为6.1×10-8mol.L-1。对浓度为5.0×10-6mol.L-1的盐酸多巴胺进行11次平行测定,其相对标准偏差为4.5(。结论:该方法简单,快速,灵敏,可用于盐酸多巴胺注射液中盐酸多巴胺含量的测定。

Objective：To develop a high sensitive and simple spectrophotometric method for the determination of dopamine hydrochloride.Methods：The determination of dopamine hydrochloride based on the inhibition of hemoglobin for the oxidation of H2O2 with acid chrome blue K（ACBK） in the NH3-NH4Cl buffer.The percentage inhibition （I﹪） of system is calculated under the optimal experimental condition.Results：The calibration curve is linear in the range of 1.1×10-7-1.1×10-5 mol/L-1,with the detection limit of 6.1×10-8 mol/L-1.The relative standard deviation of this method is 4.5% at 5.0×10-6 mol/L-1 for 11 determination.Conclusion：This method is simple,rapid and sensitive which is suitable for the determination of dopamine hydrochloride in dopamine hydrochloride injection with satisfactory results.
