摘要
目的:建立测定大鼠尿中拉米夫定浓度的毛细管电泳方法,并应用此方法研究拉米夫定经大鼠尿排泄的情况。方法:尿样经沉淀蛋白处理后,采用未涂层熔融石英毛细管柱(45 cm×75μm,有效长度为36.7 cm)分离,运行缓冲液为20 mmol.L-1磷酸盐缓冲液(pH2.5),分离电压为15 kV,检测波长为270 nm,温度为25℃。结果:测定大鼠尿中拉米夫定的线性范围为20.0~1000μg.mL-1(r=0.9994),日内和日间精密度均小于5.8%,准确度为93.8%~101.9%,提取回收率为90.3%~94.8%。大鼠灌胃给予拉米夫定后24 h内尿中原形药物的累积排泄量约占给药剂量的49.8%。结论:该方法简便、快速、经济,可用于拉米夫定在大鼠体内的排泄研究。
Objective:To develop and validate a capillary electrophoretic method for the determination of lamivudine excretion in rat urine.Method:Rat urine samples were deproteinized with acetonitrile and separated on an untreated fused silica capillary(45 cm×75 μm,effective length is 36.7 cm)with a buffer consisting of 20 mmol·L-1 phosphate buffer(pH 2.5).The applied voltage was 15 kV.The UV detection wavelength was set at 270 nm and the capillary was kept at 25 ℃.Results:The calibration curve for lamivudine was linear over the concentration range of 20.0-1000 μg·mL-1(r=0.9994).The intra- and inter-day precisions were less than 5.8% and the accuracies were 93.8%-101.9%.The extraction recoveries of lamivudine were 90.3%-94.8%.The urine cumulative excretion percentage of lamivudine within 24 h after i.g. administration of lamivudine to rats was 49.8%.Conclusion:The method is simple,rapid and economic,and suitable for the determination of lamivudine excretion in rat urine.
