Separation and purification of chlorogenic acid from Flos Lonicerae by high-speed counter current chromatography

摘要 目的:采用高速逆流色谱法对金银花提取液中的绿原酸进行分离纯化。方法:采用微波辅助提取金银花中的绿原酸,提取液经过滤、浓缩,所得浸膏作为高速逆流色谱分离的样品。采用TBE-300A型高速逆流色谱仪,以正丁醇-乙酸-水(4∶1∶5)为溶剂体系进行分离纯化,用下相作流动相,上相作固定相,流动相流速为2.0 mL.min-1,仪器转速为1000 r.min-1,温度为25℃。结果:金银花粗提物经过一步纯化,得到绿原酸的纯度为98.2%。结论:用微波辅助提取、高速逆流色谱法分离纯化金银花中的绿原酸,方法简单、有效,为绿原酸的分离纯化提供了一种新途径。 Objective:To separate and purify of chlorogenic acid from the crude extract of Flos Lonicerae by high-speed counter-current chromatography(HSCCC).Methods:Chlorogenic acid was extracted with the assistance of microwave power,then filtrated and concentrated under reduced pressure.The condensed sample was separated by HSCCC.Two-phase solvent system containing nbutanol-acetic acid-water(4 ∶1∶5)was run on a preparative scale.The lower phase was used as the mobile phase and the upper was used as the stationary phase in the head to tail elution mode.The flow rate was 2.0 mL·min-1.The running rate was 1000 r·min-1 at 25 ℃.Results:Chlorogenic acid with 98.2% purity was obtained by once separation.Conclusion:The HSCCC method is simple,high-performance and suitable for isolating and purificating chlorogenic acid from Flos Lonicerae.
