HPLC法测定烂积丸中大黄素与大黄酚的含量

Determination of Emodin and Chrysopanol in Lanji Pills by HPLC

目的建立测定烂积丸中大黄素、大黄酚含量的HPLC法。方法采用HPLC法。以SHIMADZUVP-ODS为色谱柱;流动相：甲醇-0.05%磷酸（80∶20）;检测波长：254nm;流速：1.0mL.min^-1。结果大黄素在4.304-21.52μg范围内线性关系良好,r=0.9996;大黄酚进样量在11.79-58.84 g范围内线性关系良好,r=0.9996。大黄素的加样回收率平均为102.75%,RSD为2.23%（n=6）;大黄酚的加样回收率平均为97.67%,RSD为1.66%（n=6）。结论该方法准确、灵敏度高,重现性好。

Objective To establish a HPLC method for the determination of emodin and chrysopanol in Lanji Pills.Methods Octadecylsilane bonded was used as the stationary phase,methanol-0.05% phosphate acid solution（80∶20） as the mobile phase.The column was SHIMADZU VP-ODS with detection wavelength at 254nm and the flow rate was 1.0mL· min^-1.Results The calibration curve of emodin was in good linearity over the range of 4.304～21.52μg（r=0.9996）.The calibration curve of chrysopanol was in good linearity over the range of 11.79～58.84μg（r=0.9996）.The mean recoveries of emodin and chrysopanol in Lanji Pills were 102.75%（RSD=2.23%） and 97.67%（RSD=1.66 %）.Conclusion The method is reliable,accurate and reproducible.
