摘要
目的建立人血清中7种游离脂肪酸的测定方法。方法人血清中的硬脂酸、油酸、亚油酸、十八碳三烯酸、二十碳四烯酸、二十碳五烯酸和二十二碳六烯酸通过加入正十七酸内标,经硫酸/甲醇甲酯化,正己烷萃取浓缩,气相色谱法定性、定量。结果7种脂肪酸甲酯化后可完全分离,重复性好,在5.0~500μg/mL范围内相关系数r>0.999,检出限≤1.1μg/mL,20μg/mL、100μg/mL、300μg/mL 3个浓度的加标试验,平均回收率在91.7%~98.1%之间,RSD 1.9%~4.1%之间。结论该法适用于人血清中游离脂肪酸测定。
Objective To establish method for the determination of 7 free fatty acids in human serum.Methods By adding n-Heptadecanoic acid as internal standard,Stearic Acid,oleic acid,linoleic acid,Linolenic acid, arachidonic acid,eicosapentaenoic acid and Docosahexaenoic Acid in human serum were methylesterified by H_2SO_4/methanol,extracted and concentrated by n-hexane,detected by gas chromatography.Results 7 kinds of fatty methyl ester could be separated completely after methylesterified,The linearity were ranged from 5.0~500μg/mL(r〉0.999),Minimum Limited Determination≤1.1μg/mL,The average recovery rate of 3 levels (20μg/mL,100μg/mL,300μg/mL) was91.7%~98.1%,RSD 1.9%~4.1%.Conclusion The method could be used to the determination of free fatty acids in human serum.
