茎注射外源DNA体内运输及雌不育变异株的研究 被引量:14

摘要 为探索无融合生殖基因转移途径,用穗茎注射法将无融合生殖大黍总DNA导入水稻品系桂99,获得1雌不育变异,其有性胚囊败育方式与大黍相似.为揭示穗茎注射的外源DNA在体内的运输途径,用显微放射自显影技术进行了研究.结果表明,外源DNA能经维管束的导管被运转到花器官中. In this experiment,the total DNA of Panicum maximum is injected into the stems of rice (Oryza Sativa L.) when the rice is in its early microspore developing stage.As a result,a female sterile variant (FSV 1) is identified from the recipient Gui 99 (a restorer line).The variants functional megaspore degenerates as the pattern in Panicum maximum.This aims at finding out the apomixis genes transfering way in rice.In the course of this experiment,a micro radioautographic technology is introduced into the study on the transportation way of exogenous DNA in vivo when it is injected into internodes.Results obtained show that exogenous DNA can be transported into flower organs through vessels.
