应用^(60)Coγ射线诱变选育武夷菌素高产菌的研究 被引量:9

摘要 武夷菌素是一种广谱、高效、低毒的生物农药,对蔬菜病害及果树、粮食作物病害有着明显的防治效果。但目前生产上所用的武夷菌素产生菌的效价都相对较低,为进一步提高武夷菌素产生菌药效,降低生产成本,研究以L7为出发菌株,采用3种不同剂量(500Gy,1000Gy,1500Gy)的60Coγ射线对其进行诱变处理。通过琼脂块法初筛,从3582株突变株中选育出78株相对高产菌株进行摇瓶复筛,最终选出5株效价最高的菌株(C-28,C-42,C-64,C-75,C-76)进行遗传稳定性测定。5次传代后,结果显示,菌株C-75遗传稳定性较好,能够稳定保持高效价不变(达到5 528μl/L),较出发菌株L7(效价4112μL/L)提高了34.4%。 Wuyiencin is a kind of broad spectrum,efficient and low-toxic agricultural antibiotic,which is obviously effective to the disease of vegetables,fruit trees and grain crops.At the present,however,the efficacy of Wuyiencin remains lower relatively.In order to further increase the efficacy of Wuyiencin and reduce the cost of production,this research adopt three dosages(500 Gy,1000 Gy,1500 Gy) of ^60Co γ rays to mutagenesis strain L7.78 strains are selected from 3582 mutant strains by screening of agar blocks,then ferment these 78 strains to make secondary selection.Finally,select the most effective 5 stains(C-28,C-42,C-64,C-75,C-76) from above 78 stains to test genetic stability.The result showed that,after five generations,C-75 strain well remained genetic stability;whose titer can reach 5528μl/L.The titer of C-75 strain is higher 34.4% than L7 strain,which titer is merely 4112 μl/L.
