鞘内注射PI3K特异性抑制剂LY294002对小鼠骨癌痛的影响 被引量:2

Effect of Intrathecal Injection with PI3K Specific Inhibitor(LY294002) on Mice with Bone Cancer Pain

摘要 [目的]了解鞘内注射PI3K特异性抑制剂LY294002对小鼠股骨癌痛的影响。[方法]C3H/HeJ小鼠32只,随机分为对照组、假手术组、骨癌组和鞘内LY294002+骨癌组(抑制剂组),每组8只,分别于造模前和模型建立后的第8、10、12、14、17、21d测定各小鼠机械痛敏(机械缩腿阈值,mechanical withdrawal threshold,MWT)和热痛敏(热缩腿潜伏期,thermal withdrawal latency,TWL),LY294002术前鞘内注射。[结果]与术前相比较,骨癌组小鼠术后12、14、17、21dMWT明显下降(t=4.527,P=0.003;t=3.698,P=0.008;t=4.043,P=0.005;t=4.309,P=0.004),TWL明显降低(t=5.946,P=0.001;t=5.214,P=0.001;t=6.786,P=0.000;t=5.758,P=0.001);与假手术组相比较,骨癌组小鼠术后12、14、17、21dMWT明显下降(P=0.027;P=0.000;P=0.000;P=0.000),TWL明显降低(P=0.000;P=0.000;P=0.000;P=0.000)。与骨癌组相比较,抑制剂组小鼠在12、14、17、21dMWT明显升高(P=0.031;P=0.001;P=0.002;P=0.000),TWL明显延长(P=0.002;P=0.000;P=0.000;P=0.000);与术前和对照组相比较,假手术组在各时间点的MWT和TWL均无明显变化(P>0.05)。[结论]鞘内注射PI3K特异性抑制剂LY294002能明显减轻骨癌痛小鼠机械痛敏和热痛敏,提示PI3K在脊髓水平参与了骨癌痛信息传递。 [Purposel To investigate the effect of intrathecal injection PI3K specific inhibitor(LY294002) on mice with bone cancer pain. [Methods] Thirty-two C3H /HeJ mice were randomly divided into 4 groups (n=8 each): control group, sham operation group, bone cancer group, bone cancer+LY294002 group. The mechanical withdrawal threshold (MWT) and thermal withdrawal latency (TWL) were measured at the time of pre-operation and the 8d, 10d, 12d, 14d, 17d, 21d post operation. LY294002 was intrathecal injection before operation. [Results ] Compared with pre-operation, MWF decreased significantly in bone cancer group at the 12d, 14d, 17d, 21d post operation (t=4.527,P=0.003;t=3.698,P=0.008;t= 4.043, P =0.005 ; t =4.309, P =0.004 ) ; and TWL shortened significantly ( t =5.946, P =0.001 ; t =5.214, P = 0.001;t=-6.786,P=0.000;t=5.758,P=0.001 ). Compared with sham group, MWT decreased significantly in the bone cancer group at the 12d, 14d, 17d, 21d post operation (P=0.027;P=0.000;P=0.000;P=0.000), and TWL shortened significantly too (P=0.000 ; P=0.000 ; P=0.000 ; P=0.000). Compared with bone cancer group , MWT increased significantly in bone cancer+ LY294002 group at the 12d,14d,17d,21d post oper- ation (P=0.031 ; P=0.001 ; P=0.002 ; P=0.000), and TWL prolonged significantly at the 12d, 14d, 17d,21 d after operation (P=0.002 ;P=0.000;P=0.000 ;P=0.000). Compared with pre-operation and control group, MWT, TWL in each point of time 'after operation in sham operation group did not change significantly(P〉 0.05). [Conclusion ] Intrathecal injection with PI3K specific inhibitor (LY294002 can obviously relieve heat hyperalgesia in mice with bone cancer pain. It suggests that PI3K may play an important role in the nociceptive transmission on the spinal level.
