摘要
目的检测特应性皮炎(AD)患儿外周血中辅助性T细胞亚群(Th1、Th2和Th17)数量和比例的变化,探讨其在AD发病机制中的作用。方法选择40例AD患儿,分离外周血单核细胞(PBMC),采用流式细胞术(FCM)检测Th1、Th2、Th17细胞的数量及Th1/Th2比值,用ELISA测定培养上清中IFN-γ、IL-4、IL-17水平,与同期选择的健康对照组比较并进行相关分析。结果AD患儿PBMC中CD4+IL-4(+Th2)细胞的数量、CD4+IL-17(+Th17)细胞的数量均高于健康对照组([19.56±4.9)%vs(12.91±1.7)%,P<0.05;(26.61±2.6)vs(11.4±1.2),P<0.05],Th1/Th2细胞的比值低于健康对照组([0.98±0.13)vs(1.84±1.6),P<0.05]。结论IL-17+Th17细胞与Th1/Th2比值变化可能参与AD患儿的免疫学发病机制。
Objectlve To determine the variations of Th17 cell and Th1/Th2 subsets in peripheral blood of patients with atopie dermititis (AD) and explore their significanees. Methods The levels of CD4+IFN-γT (Th1), CD4IL-4T(Th2) CD4IL-17T (Th17) subsets in peripheral blood of 40 patients with AD and 40 healthy controls were analyzed by flow cytometry with bi-eolor immunofluoreseent staining. The levels of IFN-γ,IL-4 and IL-17 were detected by ELISA. Results The levels of peripheral blood Th17 and the number of Th2 subsets were significantly increased in patients with AD in comparison with that of healthy controls [(19.56±4.9)% vs (12.91±1.7)%, P〈0.05;(26,61±2.6) vs (11.4±1.2),P〈 0.05,],without showing significant difference. Conclusion The variations of ratios of IL-17 Th17 to Th1/Th2 in peripheral blood of AD may may be associated with the pathogenesis of AD.
