高糖对体外培养的人脐静脉内皮细胞中c-fos的影响研究

摘要 目的观察高糖作用人脐静脉内皮细胞时c-fos基因mRNA和蛋白的表达的变化。方法提取并体外培养人脐静脉内皮细胞,分别加入不同浓度葡萄糖(5.5mmol/L、11mmol/L、22mmol/L、44mmol/L)并分别作用不同时间,应用逆转录聚合酶链反应(RT-PCR)检测c-fos基因mRNA的表达,免疫细胞化学检测c-fos蛋白表达。结果经高糖处理的人脐静脉内皮细胞中,c-fos基因mRNA的转录水平明显高于对照组(P<0.05或P<0.01)。但是脐静脉内皮细胞经22mmol/L葡萄糖作用1h,c-fos基因的mRNA水平不再升高,而呈下降趋势(P<0.05)。经22mmol/L葡萄糖作用2h后,c-fos蛋白表达达到最高峰。结论高糖能引起脐静脉内皮细胞c-fosmRNA和蛋白表达增高。 Objective To observe the transcript and protein expression levels of the c-fos in human umbilical vein endothelial cells cultured with high glucose in vitro. Methods Human umbilical vein endothelial cells were cultured in vitro with glucose at different concentrations or for different cultural time respectively. The transcript levels of c-fos was measured by RT-PCR. The protein expression of c-fos was detected with immunocytochemistry. Results The transcript and protein expression levels of c-fos in high glucose groups were higher than those of control group ( P 〈0. 05 or P 〈0. 01 ). Cultured with glucose at 22 mmoL/L, the transcript of c-fos mRNA increased in 1 h, the protein expression of c-los reacheol the summit aftertwo hours, and then be- gan to decrease( P 〈 0. 05 ). Conclusions The transcript and protein expression of c-fos gene increase in HU- VECs, cultured with high glucose.
