苏云金杆菌制剂毒素蛋白的SDS-PAGE-TLC定量分析

QUANTITATIVE METHOD OF TOXIC PROTEINS IN BACILLUS THURINGIENSIS PRODUCTS BY SDS-PAGE-TLC

为了更合理地评价生物农药苏云金杆菌产品的质量,研究了苏云金杆菌制剂中毒素蛋白定量测定的方法。样品经溶解后,用SDS-PAGE对毒素蛋白(130KD)进行分离,用薄层扫描仪扫描区带面积定量。该方法RSD为3.01％～5.10％,准确度为96.61％～99.74％,蛋白含量与经生物测定得到的毒力效价之间有较好的相关性,可以作为评价我国苏云金杆菌产品质量的标准化方法。

This article presents a quantitative method of toxic proteins in Bt products for evaluating the Bt products in the market. The toxic proteins are dissolved and separated by SDS-PAGE. The band of toxin protein is quantified by TLC. RSDs of the method vary from 3. 01% to 5. 10%. Its fortified recoveries vary from 96. 61% to 99. 74%. In some instances, the assay data correlate with bioassay data. This method can be used as the standard method of Bt products.
