MMP9特异性锤头状真核表达载体的构建及其对子宫内膜异位症细胞侵袭力的抑制作用 被引量:2

摘要 目的:构建针对MMP9基因的特异性锤头状核酶真核表达载体,探讨其对子宫内膜异位细胞侵袭力的抑制作用。方法:运用计算机软件人工设计并合成核酶基因,将核酶基因克隆到真核表达载体peDNA3.1中,阳性重组子由BamHI和EcoR I酶切,琼脂糖凝胶电泳鉴定证实。将重组质粒转染入子宫内膜异位细胞中,transwell小室法检测重组质粒对子宫内膜异位细胞侵袭力的抑制作用。结果:经酶切电泳证实合成的核酶基因序列正确并已被准确克隆入pcDNA3.1的BamHI和EcoR I酶切位点之间,命名为pcDNA3.1-RZ。Western blot显示转染核酶的靶细胞中MMP9蛋白的表达显著下降;transwell小室法显示pcDNA3.1-RZ可明显抑制子宫内膜异位细胞的侵袭力。结论:pcDNA3.1-RZ能明显抑制子宫内膜异位细胞的侵袭性生长,这种作用可能与MMP9蛋白的表达下调有关。
