摘要
目的研究中药复方对腹主动脉缩窄大鼠左室心肌血管紧张素Ⅱ-1型(AT1)受体mRNA表达及其受体蛋白改变的干预作用。方法银夹法建立腹主动脉缩窄大鼠模型,用免疫组化法进行心肌AT1受体蛋白染色,即时PCR扩增后分析并测定其mRNA表达,观察中药复方对大鼠左室心肌质量指数(LVMI)、心肌超微结构和心肌AT1受体蛋白与其mRNA表达改变的干预作用。结果造模8周时,LVMI、左室AT1受体蛋白及其mRNA表达方面,模型组较假手术组显著升高,中药复方组较模型组显著降低。结论腹主动脉缩窄大鼠所致心肌纤维化心肌AT1基因转录与翻译水平发生改变,通过AT1下游信号通路传导作用,促进心肌纤维化。中药复方可延缓甚至逆转心肌的纤维化进程。
Objective: To investigate the international effect of Chinese herb formula on the receptor and its mina expression left ventricular myocardium angiotensin li type - 1 in abdominal aortic constriction rats. Methods: The rats model established by the silver clip. The AT1 receptor protein were stained by immunohistocher0ical method. The mRNA expression was analysed and tested by the PCR amplification. There were some indices observed including: left ventricular mass index, myocardial infrastructure, AT1 receptor protein and its mina expression. Results: At the 8th week, the LVMI, the AT1 receptor protein and its mina expression of left ventricular myocardium were increased obviously in the model group while it compared with those in sham- operation group. However, compared with the model group, these indices were decreased significantly in the Chinese herb formula group. Conclusion: Through the conduction effect of AT1 downstream signal pathway, the myocardial fibrosis is promoted by the level variations of AT1 gene transcription and translation in abdominal aortic constriction rats.
