摘要
目的研究炎黄四虫通脉灵对截瘫大鼠肢体运动功能恢复的规律并探讨其修复机制。方法将120只SPF级健康成年SD大鼠随机分为5组,即空白对照组、模型对照组、药物对照组和炎黄四虫通脉灵大、小剂量组各24只。除空白对照组外,其余4组均于胸椎10(T10)平面切断脊髓并切除2 mm使其完全横断。于伤后24 h及8周后行后肢运动功能Tarlov评分,并于伤后8周行组织学、免疫组化、细胞凋亡程度测定。结果脊髓损伤后24 h和经治疗后第8周,模型对照组、药物对照组和炎黄四虫通脉灵小、大剂量组运动功能评分均低于空白对照组(P<0.01)。治疗后第8周,药物对照组和炎黄四虫通脉灵小、大剂量组运动功能评分均明显高于模型对照组(P<0.01),尤以炎黄四虫通脉灵大剂量组疗效好。伤后后肢运动功能均有不同程度的恢复,8周时恢复速度最明显。光镜下观察脊髓组织,炎黄四虫通脉灵大剂量组恢复最明显,可见点灶状出血、坏死,范围较局限,空泡变性较少,脊髓组织中胶质纤维酸性蛋白(GFAP)的表达量最轻,凋亡细胞最少(P<0.01)。脊髓损伤后8周,各组均可见凋亡细胞,空白对照组细胞凋亡指数(AI)与其它4组比较差异有统计学意义(P<0.05),凋亡细胞最少。与模型对照组比较,药物对照组和炎黄四虫通脉灵小、大剂量组细胞A l有统计学意义(P<0.05),凋亡细胞少。炎黄四虫通脉灵大剂量组胶质细胞和神经元都存在凋亡,但以神经元为主,白质和灰质中都存在凋亡细胞,但多数分布于灰质,较炎黄四虫通脉灵小剂量组和药物对照组轻(P<0.01)。结论炎黄四虫通脉灵可促进脊髓损伤大鼠肢体功能的恢复。
Objective To research the recovery pattern and mechanism of Yanhuang Sichong Tongmailing on complete spinal cord transection in rats. Methods 120 SPF SD rats were divided into five groups, blank control group, model control group, positive drug group, low - dose Yanhuang Sichong Tongmailing group and high - dose Yanhuang Sichong Tongmailing group. Except the blank control group, the spinal cords of other SD rats were transec- ted at Tl0 plane and a 2 mm spinal cord removed for a complete transection. The functional recovery of the hind limb was evaluated using Tarlov 24 hours and 8 weeks after injury. At the 8th week after injury, experiment was done on histology, immunohistochemistry and apoptosis degree. Results The hind limb function score in model control group, positive drug group, high - dose Yanhuang Sichong Tongmailing group and low - dose Yanhuang Sichong Tongmailing group were most obvious lower than that in blank control group ( P 〈 0. 01 ). The hind limb function score in positive drug group, high - dose Yanhuang Sichong Tongmailing group and low - dose Yanhuang Sichong Tongmailing group were higher than that in model control group at the 8th week ( P 〈0.01 ). The hind limb function of injured animals in all groups recovered at different degrees, recovery of the 8th week was most obvious. In high - dose Yanhuang Sichong Tongmailing group blood of point or flake and necrosis of was limit, few cells of degeneration, few GFAP cells in spinal cords and the fewest apoptosis in spinal cord than those in other groups( P 〈0.01 ). At the 8th week after injured AI in blank control group was the lowest in all groups ( P 〈0.05 ). AI in positive drug group, high - dose Yanhuang Sichong Tongmailing group and low - dose Yanhuang Sichong Tongmailing group were lower than that I model control group ( P 〈 0.05 ). Conclusion Yanhuang Sichong Tongmai reagent can improve recovery of body movement function of the complete spinal cord transection In rats.
