摘要
目的探讨雪芪通肾汤对糖尿病肾病大鼠肾功能及p38丝裂原活化蛋白激酶(p38MAPK)mRNA表达的影响。方法将46只大鼠随机分为4组,即正常组10只,模型组、贝那普利组和雪芪通肾组各12只。模型组、贝那普利组、雪芪通肾组用采用链脲佐菌素(STZ)造模。造模成功后,各治疗组均给药24周。取动脉血测肾功能,取左侧肾脏,检测p38MAPK mRNA在肾组织中的表达。结果贝那普利组、雪芪通肾组较模型组的血肌酐(Cr)、尿素氮(BUN)均降低(P<0.01)。雪芪通肾组较正常组Cr无明显变化(P>0.05)。半定量分析结果表明贝那普利组、雪芪通肾组p38MAPK mRNA的表达比模型组有所降低(P<0.01),比正常组有所升高(P<0.01)。结论雪芪通肾汤可降低糖尿病大鼠肾脏p38MAPK mRNA表达,保护肾功能,延缓糖尿病肾病进程。
Objective To investigate the effect of Xueqi Tongshen decoction on the expression of p38 MAPK RNA and renal function in rat with diabetic nephropathy. Methods 46 rats were randomly divided into four groups, normal group ( n = 10), model group ( n = 12), Benazepril group ( n = 12) and Xueqi Tongshen decoction group ( n = 12). Diabetic model rats induced by STZ were applied on model group, Benazepril group and Xueqi Tongshen decoction group. Rats in different groups were treated by homologous medicines. The expression of p38 MAPK RNA and renal function in rat were measured. Results Scr and BUN in Benazepril group and Xueqi Tongshen decoction group were decreased in comparison with those in model group ( P 〈0.01 ). There was no difference between normal group and Xueqi Tongshen decoction group on the levels of Scr and BUN ( P 〉 0.05 ). The expression of p38 MAPK RNA in Benazeprit group and Xueqi Tongshen decoction group was decreased in comparison with those in model group ( P 〈 0.01 ), increased in comparison with that in normal group ( P 〈 0.01 ). Conclusion Xueqi Tongshen decoction can inhibit the expression of P38 MAPK RNA in rat with diabetic nephropathy, protect kidney and delay the development of diabetic nephropathy.
