HSP70与p53蛋白在肝细胞癌组织中的表达及相关性研究

The Expression and Correlation of HSP70 and p53 Protein in Hepatocellular Carcinoma

目的探讨HSP70及p53蛋白在肝癌组织中的表达及相关性。方法采用免疫组织化学S-P法检测69例肝癌组织及其癌旁肝组织的HSP70和p53蛋白,并与10例正常肝组织进行对照分析。结果HSP70在肝癌组织、癌旁肝组织及正常肝组织中的阳性表达率分别为47.8%(33/69)、81.2%(56/69)、10.0%(1/10),其阳性表达与瘤栓形成(P<0.01)、组织分化程度(P<0.05)有关,而与性别、年龄、肿瘤大小、有无包膜无关(P>0.05);p53蛋白在肝癌组织、癌旁肝组织和正常肝组织中的阳性表达率分别为56.5%(39/69)、1.4%(1/69)、0(0/10),其阳性表达与瘤栓形成(P<0.01)、组织分化程度(P<0.05)有关,而与性别、年龄、肿瘤大小、有无包膜无关(P>0.05);HSP70与p53蛋白在肝癌组织中的表达呈正相关(r=0.460,P<0.05)。结论HSP70、p53蛋白在肝癌组织中高表达,可能参与了HCC的发生、发展,且具有协同作用,两者可作为肿瘤侵袭性生物学行为的预测指标。

Objective To study the expression and correlation of HSP70 and p53 protein in hepatocellular carcinoma. Methods The immunohistochemistry S-P method was used to detect the expression of HSP70 and p53 protein in 69 cases with HCC and its paraneoplastic tissues, 10 cases of normal liver tissues were used as control group. Results The positive expression rates of HSP70 were 47.8% （33/69）, 81.2% （56/69）, 10.0% （1/10） in HCC, paraneoplastic tissues and normal liver tissues respectively. There was a positive correlation between HSP70 and tumor thrombus （ P 〈 0.01 ） and the degree of tumor differentiation（ P 〈 0.05 ）. No correlation was found between HSP70 expression and clinicopathologic characteristics such as gender, age, tumor size and tumor capsular（P 〉 0.05 ）. The positive expression rates of p53 protein were 56.5% （39/69） , 1.4% （1/69） in HCC, paraneoplastic tissues respectively. The positive expression of p53 protein was not found in normal liver tissues. There was a positive correlation between p53 protein and tumor thrombus （P 〈 0.01 ） and the degree of tumor differentiation （ P 〈 O. 05 ）. No correlation was found between p53 protein expression and clinicopathologic characteristics such as gender, age, tumor size and tumor capsular（ P 〉 0.05 ）. There was positive correlation between HSP70 and p53 protein in HCC （r = 0. 460, P 〈 0.05）. Conclusion The overexpression of HSP70 and p53 protein may be involved in the occurrence and development of HCC. There was the synergistic effect between the two. HSP70 and p53 protein may serve as predictors for determining tumor aggressive behavior in HCC.
