甘啤4号大麦幼胚愈伤组织诱导及植株再生的研究 被引量:5

摘要 本研究以甘啤4号的幼胚为外植体材料,以模式品种Golden promise为对照,研究了不同培养基和激素配比对甘啤4号幼胚愈伤组织的诱导及植株再生的影响。研究结果表明,在同一培养基上,甘啤4号出愈率低于Golden promise,但都达到50%以上;在诱导愈伤组织的过程中,甘啤4号在IM2培养基上的出愈率较高,达71%,IM1与IM2培养基上形成的愈伤质量较IM3好,说明培养基对愈伤组织的出愈率起决定作用;且研究发现dicamba较2,4-D能提高大麦幼胚的出愈率。在相同的培养基上,Golden promise的诱导率明显高于甘啤4号,可见基因型对大麦愈伤组织的诱导率起决定作用。且不同的培养基和不同激素配比对幼胚愈伤组织的诱导率及分化率不同,分化培养基DM1的分化率高于DM2,这说明除基因型外,激素也对植株再生产生重要影响。研究证明甘啤4号愈伤组织诱导率及分化率均较高,适合于作为遗传转化的受体材料。 In this research, we employed the immature embryos from Ganpi4 Hao and Golden promise, a beer barley vareity in Gansu province and a model baeley cultivar, as the explant material and the referrence, respectively, in order to study the effects of different medium and the matching of hormones on callus induction of Ganpi4 Hao. The result showed that the callus initiating rate of Ganpi4 Hao was lower than Golden promise, but both of them reached 50% above. During the induction of callus, the callus initiating rate of Ganpi4 Hao was higher in IM2 medium, which was up to 71% and the quality of callus from IM1 and IM2 was better than that from IM3, the result indicated that medium played a decisive part in the callus induction rate. Further research found that dicamba can increase the callus initiating rate of the immature embryos obviously to 2,4-D. In the same medium, the induction rate of Golden promise displayed significantly higher than Ganpi4 Hao which demonsrated that genotype set a decisive role in the induction rate of barley callus. And we still found that different media and the matching of hormones had different effects on induction rate and differentiation rate of barley callus and the differentiation of DM1 was higher than DM2, which indicated that apart from genotype, hormone also had an important impact on plant regeneration. The research proved that Ganpi4 Hao was identified with higher performance both in induction of embryonic callus and plant regeneration and was suitable as the genetic transformation recipient material.
