摘要
狼毒大戟为大戟科多年生草本植物。本研究以狼毒大戟(Euphorbia fischeriana)为试验材料,构建狼毒大戟cDNA文库及EST测序。研究结果表明,我们成功地构建了狼毒大戟的cDNA文库,并获得了2099个EST的序列。进一步对这些ESTs数据进行了Unigene的拼接、ORF预测和相应的功能注释及分类,发现了一些可能参与Prostratin生物合成的候选结构基因Transketolase,NADPH-cytochrome P450reductase,Cy-tochrome P450,S-adenosylmethionine synthetase,acetyltransferas,1-deoxy-D-xylulose-5-phosphate synthase(DXP)和候选调节基因AP2类ERF转录因子(AP2-like ethylene-responsive transcription factor,AP2-like ERF),此研究为今后开展抗HIV次生代谢物Prostratin的代谢工程奠定了基础。
Euphorbiafischeriana is a perennial herbaceous plant of Euphorbiaceae. In this research, we employed Euphorbiafischeriana to be as materials for constructing a cDNA library, and then sequencing acquired EST. Here we presented that we sucessfully constructed a cDNA library of Euphorbiafischeriana with sequenced 2099 ESTs. Further we analyzed these ESTs datas by Unigene assembly, ORF prediction, funtional annotation and classifica- tion, we found some candidate structural genes, which might be involved in biosynthesis of Prostratin, such as, Transketolase, NADPH--cytochrome P450 reductase, Cytochrome P450, S-adenosylmethionine synthetase, acetyltransferas, 1-deoxy-D-xylulose-5-phosphate synthase (DXP), and candidate regulatory genes AP2-1ike ERF. This work would provide much more EST information for developing Prostratin metabolic engineering in the future.
