摘要 表面增强激光解析离子化飞行时间质谱技术是新一代蛋白质组研究技术,它使用特殊的增强表面直接捕获待测样本中的蛋白质分子,能够快速、敏感地发现并鉴定蛋白,提供一个高通量和高灵敏度的检测平台。最初主要应用于疾病早期诊断相关的蛋白质标志物。目前在预测肿瘤化疗敏感性方面的应用也逐渐增多。本文除对表面增强激光解析离子化飞行时间质谱技术做详尽的介绍外,重点阐述其在预测肿瘤化疗敏感性方面的应用,并对其优缺点和发展前景进行评估。 Surface-enhanced laser desorption/ionization-time of flight-mass spectrometry( SELDI-TOF- MS) is a new proteomic technology. It can use special surface to capture the protein of sample, and indentify the protein rapidly and sensitively. It is a high-throughput and high-sensitive platform. It was mainly used to find the biomarkers in diseases for early diagnosis. Currently, it is gradually used to predict the chemosensitivity of tumor treatments. In this article, the development and technology of SELDI-TOF-MS are introduced, the appraisals on its pros and cons, as well as the future prospective is also expounded.
