摘要
目的探讨茶多酚对前列腺癌PC-3M细胞增殖及凋亡的影响。方法设立对照组与实验组,采用MTT比色法检测TP对PC-3M细胞增殖的影响;吖啶橙/溴化乙锭染色法观察诱导细胞凋亡情况;流式细胞术分析细胞周期;RT-PCR和Western blot检测Caspase-3基因的转录与表达。结果与对照组比较,MTT法示实验组对PC-3M细胞的增殖抑制作用增强;细胞染色法示实验组凋亡率增高;流式细胞术显示细胞阻滞于G1期,增殖指数降低;RT-PCR与Western blot示Caspase-3基因转录与翻译上调,上述结果在一定范围内呈时间、剂量依赖性(P<0.05)。结论茶多酚对人前列腺癌PC-3M细胞具有增殖抑制、促进凋亡作用,与上调caspase-3基因的转录与表达有关。
Objective To investigate the regulatory effect of tea polyphenol on proliferation inhibition and apoptesis of inhuman prostate cancer PC-3M cells. Methods The PC-3M cells were divided into treatment groups and control group. The inhibition of cell proliferation was detected by MTT assay, the cell cycle phase was analyzed by flow cytometry, the apoptosis was inspected by acridine orange/ethidium bromide fluorescent staining method, and the transcription and translation of caspase-3 in carcinoma cells was observed by RT-PCR and western blot. Results After being treated with tea polyphenol,the PC-3M cell growth was inhibited, so the rate of proliferation of PC-3M cells decreased. Partial cells presented the morphological change of apoptosis under the fluorescent microscope, and the apoptotic rates of the treatment groups increased respectively. The cells in S and G2/M phase increased,the cells in G0/G1 phase was decreased. With the increasing of concentration of tea polyphenol,the caspase-3 expression was also increased. The results presented was shown as a time-and dose-dependent manner in a certain range. Condusion Tea polyphenol could inhibit the growth of PC-3M cells and induce ~e apoptosis, the signal transduction pathway of up-regulation of caspase-3 may one of its molecular mechanisms.
