摘要
目的观察位于染色体22q12上的THOC5和ADRBK2基因在人脑胶质瘤肿瘤组织中mRNA表达。方法应用逆转录-聚合酶链反应(RT-PCR)及图像分析方法,检测17例人脑胶质瘤肿瘤组织和8例正常人脑组织中THOC5和ADRBK2基因mRNA表达。基因表达水平用THOC5或ADRBK2基因/β-肌动蛋白(β-actin)灰度比值×100表示。结果ADRBK2基因在正常人脑组织和人脑胶质瘤肿瘤组织中,均存在弱表达,统计分析显示无显著性差异(P>0.05);THOC5基因在正常人脑组织中弱表达,在人脑胶质瘤肿瘤组织中高表达。统计分析显示:人脑胶质瘤肿瘤组织中THOC5基因mRNA表达与正常人脑组织中THOC5基因mRNA表达比较,有显著性差异(P<0.05)。结论THOC5基因高表达可能与胶质瘤的发生、发展相关。
Objective To obserev the mRNA expression of THOC5 and ADRBK2 genes in human gliomas. Methods The mRNA expression of THOC5 and ADRBK2 genes in 17 cases gliomas and 8 cases normal brain tissues were detected by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) in the presence of specific primers. The level of THOC5 and ADRBK2 gene expression were expressed as ratio expression rate (RER) of THOC5 or ADRBK2 gene/β-actin 100. Results ADRBK2 gene had weak expression in normal brain tissues and gliomas, and there was no significanted difference( P 〉 0.05) ;THOC5 gene had weak expression in normal brain tissues, had significantic expression in gliomas. There was significanted difference of THOC5 gene between normal brain tissues and gliomas( P 〈 0.05). Conclusion It can be inferred that the overexpression of THOC5 gene may be involved in glioma orgin.
