摘要 目的研究细胞凋亡相关因子在胃肠道间质瘤中的表达及其意义。方法RT-PCR检测NF-κB、XIAP、bcl-2与survivin在124例GIST中的mRNA水平。结果在GIST中,NF-κB mRNA随NIH分级改变而表达上调(P<0.05),并随肿瘤恶性度增加,临床分期越晚而增加程度越重,与肿瘤的侵袭转移、黏膜受侵密切相关,而与年龄、性别、组织学类型无关。XIAP mRNA随NIH分级表达上调(P<0.05);XIAP表达与GIST组织分化程度呈负相关,即分化程度越低,XIAP阳性表达率愈高,伴有肿瘤侵袭转移的GIST病例XIAP表达水平较高,同时XIAP的表达与黏膜受侵密切相关,而与年龄、性别、组织学类型无关。bcl-2 mRNA水平表达上调,与NIH分级、黏膜受浸有关,而与年龄、性别、组织学类型、转移复发及周围组织浸润无关。Survivin mRNA在GIST中的表达与NIH分级、肿瘤侵袭转移、黏膜受侵等因素有关,而其表达与年龄、性别以及组织学类型无关。由于NF-κB的调控作用,各指标之间mRNA的水平变化呈正相关。结论GIST中NF-κB、XIAP、bcl-2与survivin有望作为判断GIST转移及预后的参考指标。 Objective To investigate the detection and significance of apoptosis related factors in gastrointestinal stromal tumors. Methods The expression of NF-κB,XIAP,bcl-2 and survivin were detected with RT-PCR using specimens from 124 cases of GIST. Results In gastrointestinal stromal tumors the mRNA expression of NF-κB, XIAP, bcl-2 and survivin is closely related to NIH clas- sification, mucosal invasion and lymphatic metastasis( P 〈 0.05). They have no relationship with age,sex and histological type( P 〉 0.05). Based on the regulation of NF-κB regulation, the expression of them are of positive Correlation. Conclusion The expression of NF-κB,XIAP,bcl-2 and survivin have relationship with the prognosis of GIST.
