摘要
目的利用荧光定量PCR检测健康育龄妇女阴道中的主要乳酸杆菌,并比较不同季节阴道乳酸杆菌分离率及数量的差异。方法分别在春夏季、秋冬季采集健康妇女阴道分泌物标本97例(春夏季63例、秋冬季34例),应用SYBR GreenI荧光定量PCR方法测定标本中5种乳酸杆菌16SrRNA基因拷贝数量并比较两组间的差异。结果健康妇女阴道乳酸杆菌的分离率达到96.9%,其中卷曲乳酸杆菌分离率66.0%、惰性乳酸杆菌45.4%。健康妇女阴道乳酸杆菌数量(Log10 copies/ml)为6.21±1.28,其中卷曲乳酸杆菌数量最多,为5.09±2.98。春夏季与秋冬季比较,阴道卷曲乳酸杆菌的数量显著增加(P<0.05)。结论卷曲乳酸杆菌和惰性乳酸杆菌是健康妇女阴道中的主要乳酸杆菌,卷曲乳酸杆菌数量占绝对优势。阴道中的卷曲乳酸杆菌数量在不同季节存在明显差异。
Objective To study the seasonal variation of vaginal lactobacilli in based on the real-time fluorescent quantitative PCR.Metilods Collecting 97 vaginal-fluid samples of beating age women in difference seasons(63 in spring and summer,34 in autumn and winter). Determining the prevalence of 5 lactobacillus species predominate in the women vagina. Real-time PCR procedures' based on 16SrRNA gene sequences specific for Lactobacillus were developed and used for quantitative detection. Results A total of 94 (96.9%) of 97 vaginal samples were positive for lactobacillus. L. crispatus was the most frequent isolated Laetobaeillus species from the vagina (66.0%) of the subjects, followed by L. iners (45.4 % ). log10 laetobacillus counts is 6.21 ± 1.28, log10 L. crispatus counts is 5.09 ± 2.98. A significant increasing was in the quantity of L. crisp between the summer group and the winter group. Conclusion L. crispatus and L. iner are the most common species of vaginal lactobacilli. The number of L. crispatus in vagina is significantly different between summer and winter.
