Extraction and Separation and Purification of Estradiol from Chestnut Pollen

摘要 对板栗花粉雌二醇提取条件进行优化,雌二醇的最佳提取条件为:提取温度70℃、提取时间2.5h、乙醇浓度80%、液固比10:1(V/V),用HPLC定量雌二醇的含量为9.72μg/g。AB-8大孔树脂柱层析分离纯化雌二醇,薄板层析和HPLC表征分离纯化情况,其理想分离纯化体系为:洗脱液60%乙醇4BV、丙酮4BV依次洗脱。 Extraction conditions of estradiol from chestnut pollen are studied, the optimal extraction conditions of estradiol are as follows: temperature of 70 ℃, time of 2.5 h, ethanol content of 80% and ratio of solid to liquid of 1:10(V/V), the content of estradiol in the extracts is 9.72 μg/g by HPLC quantitative determination. Separation and purification of estradiol using AB-8 macroporous resin column chromatography, throughing the TLC and HPLC characterization, the perfect separation and purification system are as follows: the resin column is eluted with 4 BV ethanol content of 60% and 4 BV acetone successively.
