摘要
目的:观察丹酚酸胶囊对Beagle犬急性心肌缺血模型缺血心肌的保护作用。方法:连续2周每日给予Beagle犬25mg.kg-1、50mg.kg-1、100mg.kg-1丹酚酸胶囊后,观察二步法结扎冠状动脉致Beagle犬急性心肌缺血模型的心率(HR)、血压(MAP)、心肌血流量(MBF)和冠脉阻力(CVR)的变化,以及血清肌酸激酶同工酶(CK-MB)、丙二醛(MDA)含量、过氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-PX)活性的变化。结果给予高、中、低剂量丹酚酸胶囊组可明显降低HR、CVR、CK-MB和MDA含量(P<0.05,P<0.01),显著提高MBF、SOD活性和GSH-PX活性(P<0.05,P<0.01),对血压无明显影响(P>0.05)。结论:丹酚酸胶囊可有效降低Beagle犬急性心肌缺血程度,改善心肌供血,提高其自由基清除能力和抗氧化能力,对犬的缺血心肌具有保护作用。
Objective:To explore the effect of salvianolic capsule on acute myocardial ischemia in Beagle dogs.Methods:The left anterior descending coronary artery was ligated to make a model of acute myocardial ischemia in Beagle dogs after treatment with ig salvianolic capsule at 25mg·kg-1,50mg·kg-1 and 100mg·kg-1 once day for 2 weeks.The change of heart rate(HR),mean arterial pressure(MAP),myocardial blood flow(MBF) and coronary vascular resistance(CVR) were observed,and the serum mb isoenzyme of creatine kinase(CK-MB) activity,malondialdehyde(MDA) content,the superoxide dismutase(SOD) and glutathione peroxidase(GSH-PX) activities were detected.Results:The MBF,SOD and GSH-PX activities of salvianolic capsule treated group were significanly higher than that of model group.The HR,CVR,CK-MB and MDA level of salvianolic capsule treated group were significant1y lower than those of model group(P0.05,P0.01),and without affecting blood pressure(P0.05).Conclusion:Salvianolic capsule can protect ischemic myocardium in Beagle dogs.
