摘要
目的探讨高温和内毒素(LPS)复合因素对RAW 264.7细胞IL-1βmRNA表达的影响。方法采用体外培养的巨噬细胞系RAW 264.7细胞,分为37,40℃对照组,37,40℃LPS组。用RT-PCR方法检测RAW 264.7细胞IL-1βmRNA表达。结果RAW 264.7细胞在37和40℃加或不加LPS条件下随着时间的变化IL-1βmRNA表达各不相同,在37℃条件下,LPS组随着时间的不断延长,IL-1βmRNA表达不断增强;40℃条件下对照组IL-1βmRNA表达亦表现出不断增强,而LPS组IL-1βmRNA表达表现为先增强后减弱。结论高温和内毒素复合因素能够下调RAW 264.7细胞IL-1βmRNA表达,考虑其原因与高温和内毒素复合因素诱导产生的热休克蛋白及该条件下RAW264.7细胞的大量凋亡有关。
Objective To study the effects of high temperature and LPS on IL-1 βmRNA expression in RAW 264.7 cells. Methods Macrophage cell line RAW 264. 7 in vitro was divided into 4 groups ( control group of 37 ℃, LPS group of 37 ℃ ,control group of 40 ℃ ,LPS group of 40 ℃ ). The expression of IL-1β mRNA was detected with RT-PCR. Results The IL-1β mRNA expression of RAW 264.7 cells was different under the condition of 37 ℃ and 40 ℃ with or without LPS. The IL-1β mRNA expression of LPS group increased at 37 ℃ ; the blank group of 40 ℃ tended to ascend gradually while the LPS group of 40 ℃ ascended firstly but descended finally. Conclusion Temperature could affect the IL-1β mRNA expression in RAW 264. 7 cells. High temperature and LPS could down-regulate IL-1β mRNA expression.
